张蕾,理学博士,博士生导师,现任中国科学院福建物质结构研究所研究员,2022年入选中国科学院特聘骨干人才。在分子生物学、基因组学、蛋白质功能、生物医用材料等研究领域具有丰富的科研经历和研究基础。1996-2007年于华中农业大学农业微生物国家重点实验室完成本科、硕博研究生学历;2007-2010年在武汉大学生物医用高分子材料教育部重点实验室从事博士后研究,2010年获得武汉大学优秀博士后奖;随后在厦门大学固体表面物理化学国家重点实验室从事科研工作;2012年至今,在中国科学院福建物质结构研究所从事离子通道与肿瘤相关蛋白的结构生物学研究和生物分子靶向纳米运载体的研发,主持了两项国家自然科学基金项目和两项福建省科技项目。2017年至2018年访问学者,从事逆转肿瘤多药耐药性(MDR)的机制及靶向MDR肿瘤纳米药物研究。主要成果包括:通过小分子抑制剂的胞内靶向封闭及CRISPR/Cas9 基因敲除证明了磷酸肌醇三激酶 PI3K 110α/110β为逆转肿瘤多药耐药性的潜在靶点、PI3K-CDK6为逆转P-糖蛋白介导的肿瘤多药耐药性的信号通路轴;在此基础上设计了一种具有肿瘤靶向性的多功能纳米体系PBDF,该体系以金核枝接PLGA-SH包裹的阿霉素(DOX)和BAY-1082439颗粒,利用叶酸介导靶向肿瘤作用,有望克服P-糖蛋白介导的肿瘤多药耐药性;构建了pH敏感的“PMMMA/PLGA/抗原”口服疫苗体系,该体系在酸性的胃及多种酶存在的小肠内提供对抗原的保护作用,大肠中能缓释抗原至免疫相关的细胞,实现抗原呈递和诱导抗体形成;基于肌醇等生物可利用或生物可降解分子构建了系列高效进入动物细胞、靶向肿瘤细胞、或促进基因转染及表达的系列高效低毒副作用的药物传递体系;构建了胞内抗体的展示与筛选系统;证明了苏云金芽胞杆菌CTC菌株的晶体蛋白与表面S-层蛋白同为一种蛋白,为芽胞杆菌分类提出新的依据。在Molecular Cancer、Acta Pharmaceutica Sinica BACS AMI、Biomaterials、Journal of Materials Chemistry B、Drug Discovery Today、International Journal of Nanomedicine、Colloid surface B、Applied Microbiology and Biotechnology、Journal of Applied Microbiology 等国内外权威刊物上以第一作者和通讯作者发表了多篇学术论文。获得第十五届福建省自然科学优秀学术论文一等奖和三项国家发明专利。
